چکیده

BACKGROUND: Uridine diphosphate (UDP) is an extracellular nucleotide signaling molecule implicated in diverse biological processes via specific activation of pyrimidinergic receptor P2Y, G Protein-Coupled, 6 (P2Y6). There very little knowledge about the function and mechanism UDP rheumatoid arthritis (RA). METHODS: This study used a quasi-targeted liquid chromatography-mass spectrometry (LC-MS) approach to investigate unique expression metabolites RA synovial fluids (SF) ( n = 10) with samples from osteoarthritis (OA) as controls 10). fibroblast-like synoviocytes (FLSs) were collected tissues 5) cultured or MRS2578, P2Y6 antagonist, FLSs OA 5). Rats collagen-induced (CIA) injected UDP, MRS2578 both 9 for each group). was examined using real-time PCR, Western blotting immunohistochemistry. Cell proliferation, apoptosis migration measured CCK-8 assay, cell analysis, flow cytometry, wound healing assay Transwell respectively. The levels culture medium, fluid 36) peripheral blood CIA rats Transcreener Assay. Levels proinflammatory cytokines assay. Interleukin-6 (IL-6) ELISA flow. RESULTS: LC-MS analysis detected significantly increased SF compared SF, level positively correlated anticyclic citrullinated peptide (anti-CCP) factor (RF)levels RA. concentration verified patients healthy volunteers, stimulated IL-6 secretion inhibited their culture, these effects UDP. injection accelerated production rather than other rat model, but simultaneous suppressed alleviated CIA. tissues. CONCLUSION: These results suggest that highly expressed stimulates pathogenesis by promoting activities increase production.

Chemotherapy-induced uridine diphosphate release promotes breast cancer metastasis through P2Y6 activation

Although purinergic signaling is important in regulation of immune responses, the therapeutic potential of it in the tumor microenvironment is little defined. In this study, we demonstrate that UDP/P2Y6 signaling facilitates breast cancer metastasis both in vitro and in vivo. We found that P2Y6 is not only aberrantly expressed and mutated in most tumor types, but also highly correlated with poo...

Enzymatic Synthesis of Uridine Diphosphate Xylose and Uridine Diphosphate Arabinose.

It has been shown previously' that mung bean seedlings contain a mixture of sugar nucleotides consisting of UDPG,2 UDPGal, UDPXy, and UDPAr. In the present communication evidence is presented that these bean seedlings contain an enzymatic system capable of catalyzing the reversible formation of UDPXy and UDPAr from UTP and a-D-Xy-l-P. When a-D-Xy-l-P and UTP were incubated in the presence of an...

Uridine Diphosphate Glucose Dehydrogenase

J-Hydroxyuridine cliphosphate glucose was prepared enzymatically and 5,Bdihydrouridine diphosphate glucose was prepared by the catalytic reduction of UDP-glucose. The effectiveness of these cofactor analogues as substrates for UDP-glucose dehydrogenase was determined. The rate of oxidation of 5-hydroxyuridine diphosphate glucose was onesixth that of UDP-glucose at pH 8.7 and one-half that of UD...

Preparation of uridine diphosphate-N-acetylgalactosamine from uridine diphosphate-N-acetylglucosamine by using microbial enzymes.

A method was developed for the large scale preparation of uridine diphosphate-N-acetylgalactosamine (UDP-GalNAc) from uridine diphosphate-N-acetylglucosamine (UDP-GlcNAc) by means of microbial enzymes. With Bacillus subtilis cell-free extract as a source of UDP-GlcNAc 4-epimerase, about 35% of the UDP-GlcNAc added was converted to UDP-GalNAc. After the residual UDP-GlcNAc was degraded to uridin...

Isolation of uridine diphosphate N-acetylglucosamine and uridine diphosphate glucuronic acid from mung bean seedlings.

It has been shown previously (1) that mung bean seedlings (Phaseolus aureus) contain uridine diphosphate derivatives of n-glucose, n-galactose, n-xylose, and L-arabinose. These seedlings, as well as a number of other higher plants, also contain enzymatic systems capable of catalyzing the reversible formation of sugar nucleotides from UTPI and various monosaccharide l-phosphates (2, 3) according...
